Multiplex Assay Overview:

The assay service provided by Eve Technologies on their Human Soluble Cytokine Receptor Array facilitates advanced inflammatory response research for almost all disease states. This Discovery Assay® measures 14 cytokine receptor biomarkers simultaneously in a single microwell: sCD30, sEGFR, sgp130, sIL-1RI, sIL-1RII, sIL-2Rα, sIL-4R, sIL-6R, sRAGE, sTNFRI, sTNFRII, sVEGFR1, sVEGFR2, sVEGFR3. The assay is designed for protein analysis in serum, plasma, cell culture, and tissue homogenate. Other sample types may also be tested.

Background Information:

Cytokines play a fundamental role in the pathogenesis of many diseases and are key targets for drug development. Like cytokines, soluble cytokine receptors are involved in regulating inflammatory responses and modulating immune events. This has stimulated significant research interest in their potential role as biomarkers for various diseases and immunotherapeutic agents. Soluble cytokine receptors naturally arise from genes encoding membrane- bound receptors or are direct derivatives of the receptors themselves. Many of these soluble cytokine receptors have the ability to inhibit the binding and biological activity of their cytokine ligands, making them very specific cytokine antagonists.

The assay service provided on this array is designed for the simultaneous analysis of multiple cytokine receptor biomarkers, supplying you with quantified results for the study of inflammatory and immune responses in serum, plasma, cell culture and tissue homogenate.

Intended Sample Type:

Human serum or plasma (EDTA recommended), cell culture, and tissue homogenate. A pilot run is recommended for cell culture, tissue homogenate and other sample types not listed.

Special Sample Preparation:

Serum and Plasma samples will be diluted 1:5 by Eve Technologies to bring protein concentrations within the standard curve range, as per the manufacturers instructions for use.

Detection Method:

Luminex xMAP technology is used for multiplexed quantification of 14 Human cytokines, chemokines, and growth factors.

The multiplexing analysis is performed using the Luminex™ 200 system (Luminex, Austin, TX, USA) by Eve Technologies Corp. (Calgary, Alberta). Fourteen markers are simultaneously measured in the samples using Eve Technologies’ Human Soluble Cytokine Receptor 14-Plex Discovery Assay® (MilliporeSigma, Burlington, Massachusetts,USA) according to the manufacturer’s protocol.  The 14-plex consists of sCD30, sEGFR, sgp130, sIL-1RI, sIL-1RII, sIL-2Rα, sIL-4R, sIL-6R, sRAGE, sTNFRI, sTNFRII, sVEGFR1, sVEGFR2, sVEGFR3.  Assay sensitivities of these markers range from 3 – 115 pg/mL for the 14-plex.  Individual analyte sensitivity values are available in the MilliporeSigma MILLIPLEX® MAPprotocol or in the table below.

*Note: %CV is an appropriate measure of quality only for sample that have significant detectable concentrations for a particular analyte. Low concentration / low signal samples can often have a much higher %CV as the signals are at the limitations of the assay and fall within a flat portion of the curve.

% recovery is comparing the observed concentration to the expected concentration of samples spiked with the standard curve cocktail at various concentrations.
